But scientists are increasingly looking somewhere less obvious: your gut.
Inside your digestive system lives a dense microbial ecosystem that helps break down food, trains the immune system, produces chemical by-products and sends signals to the brain. This is known as the microbiota-gut-brain axis, and it has become one of the most serious areas of mood and mental well-being research over the past decade.
The gut is not “controlling your mind,” despite what the wellness internet often claims.
The truth is, the gut and brain are in constant biological conversation. That conversation runs through the vagus nerve, immune messengers, hormones, inflammation, microbial metabolites and pathways linked to serotonin.
And the serotonin part is where most people get the story dangerously wrong.
Around 90% of the body’s serotonin is made in the gut — but gut-made serotonin does not simply travel into the brain and make you happy.
This explains both why the gut-brain connection matters and why the hype around it can go too far.
A healthier gut environment may help support the signals your brain receives every day. But no probiotic, powder or capsule should be sold as a shortcut to mental health.
The truth is more useful than that. Your gut may not be the whole story behind your mood, but it could be one of the foundations your mood is standing on.
For years, digestion was treated like a mechanical process. Food went in, nutrients were absorbed, waste came out. Simple.
That view now looks outdated.
In a 2011 review in Nature Reviews Neuroscience, UCLA researcher Emeran Mayer described gut-brain communication as a complex, bidirectional system that helps regulate digestion and may also affect motivation, emotional state and higher cognitive function.
Bidirectional is the word that matters. Stress can disturb the gut. Gut irritation can send stress-like signals back toward the brain. Poor sleep can alter microbial balance, while an inflamed or disrupted gut environment may influence immune and nervous system pathways involved in how steady, energised or emotionally resilient someone feels.
This is why people often notice digestive changes under pressure. The nervous stomach before a presentation. The bathroom urgency before a race. The appetite swings after a rough night. These are not personality flaws; they are examples of the gut and brain reacting as one connected system.
A real-world example makes this easier to see. Imagine someone training hard, eating on the run, sleeping five hours a night and using coffee to drag themselves through the day. Their mood might feel like a mental problem, but their biology is being pushed from every direction: stress hormones are elevated, digestion is rushed, fibre intake is low, and the gut microbiome is getting less of what it needs to produce helpful metabolites.
That person may not need a motivational quote. They may need a better foundation.
Serotonin has been marketed as the “happy chemical,” which is catchy but incomplete.
In the brain, serotonin is involved in mood, sleep, appetite and emotional processing. In the gut, serotonin has different primary jobs, including helping regulate gut movement, secretion and sensation.
The shocking part is that most serotonin is produced outside the brain. The important caveat is that serotonin made in the gut does not freely cross the blood-brain barrier.
That means the common wellness claim, “fix your gut, and you’ll directly boost brain serotonin”, is not accurate.
The more accurate version is still fascinating: gut microbes may influence serotonin-related biology indirectly by affecting tryptophan metabolism, immune activity, inflammatory signalling and microbial compounds that interact with the nervous system.
One of the landmark studies here is Jessica Yano and colleagues’ 2015 paper in Cell, “Indigenous Bacteria from the Gut Microbiota Regulate Host Serotonin Biosynthesis.” The researchers found that spore-forming bacteria from the gut microbiota promoted serotonin production in colonic enterochromaffin cells and influenced gut motility and platelet function.
That study was not a mood study. It did not show that a probiotic makes people happier. What it did show was more foundational: gut bacteria can help regulate host serotonin production in the gastrointestinal tract.
Another important study, published by Gerard Clarke and colleagues in Molecular Psychiatry in 2013, found that the early-life microbiome regulated the hippocampal serotonergic system in mice in a sex-dependent manner.
Again, this is not proof that a supplement changes your personality. It is evidence that the microbiome can influence biological systems connected to brain development and serotonin signalling.
The gut-brain connection is not simple. That is exactly why it matters.
Probiotics sit right in the middle of this conversation, and they are often oversold.
Some strains have evidence for digestive support. Some are being studied for mood-related outcomes. But “probiotic” is not one thing. It is a broad category, and different strains can behave differently.
A better question is not, “Do probiotics work?”
It is: which strain, at what dose, in which person, for what outcome, and compared with what?
In 2013, Kirsten Tillisch and colleagues published a human study in Gastroenterology called “Consumption of Fermented Milk Product With Probiotic Modulates Brain Activity.” Healthy women were assigned to consume a fermented milk product with probiotics, a non-fermented milk product, or no intervention twice daily for four weeks. Brain imaging showed differences in responses to an emotional attention task and resting-state brain activity after the probiotic fermented milk intervention.
That is the kind of study that should make people curious, not reckless. It was small. It was not a depression treatment trial. But it showed that changing the gut environment could be associated with measurable changes in brain activity in humans.
In 2018, Qin Xiang Ng and colleagues published a meta-analysis in the Journal of Affective Disorders reviewing 10 clinical trials with 1,349 participants. The authors found probiotic supplementation was associated with an improvement in depressive symptoms compared with placebo, while also noting the need for more high-quality trials.
That is a promising signal. It is not permitted to claim that probiotics treat depression.
For a premium supplement brand, the responsible message is clear: probiotics may support gut conditions linked to mood biology, but they do not replace therapy, medication, medical advice, sunlight, sleep, movement or human connection.
The science is strongest when it is not exaggerated.
